I'll be honest: I mostly lucked my way into the Keeper unlocks with Head of the Keeper/Swallowed Penny combos. But the Lost is actually a very fair challenge now, with the Holy Mantle unlocked. I play him for fun sometimes, even!

The secret of defeating the Binding of Isaac? Besides Tenacity and patience to die a lot, I guess: don't go in stores with less than 15 coins, bomb blue rocks, always give money to beggars.
